双翅目(そうしもく)(ハエやカ)の唾腺染色体(だせんせんしょくたい)とは何か?

唾液腺染色体(だえきせんせんしょくたい 英語:salivary gland chromosome

双翅(そうし)類昆虫幼虫期における唾液腺の細胞核中にみられる巨大な染色体をいう。唾腺染色体、巨大染色体ともいう。一般に双翅類昆虫の唾液腺細胞は通常の細胞分裂をせずに核内分裂のみ行って巨大化する。すなわち、この細胞は休止核の状態で染色体が核内分裂を繰り返し、長大な染色体となる。個々の染色体はファイバー状になり、染色粒(クロモメアー)が同一場所に横に連なって存在するので、その部分がバンド模様に現れる。ショウジョウバエを用いた遺伝の交配実験から唾液腺染色体のバンド模様と遺伝子の関係が明らかにされ、唾液腺染色体地図が作成されている。核の中では相同染色体が2本ずつ対(つい)になってくっついているので唾液腺染色体は体細胞の染色体数の半分しかみられない。たとえば、キイロショウジョウバエでは4本(2n=8)メスアカケバエは5本(2n=10)、カは3本(2n=6)。(コトバンク

 

多糸染色体(英:polytene chromosome)

Polytene Chromosomes (多糸染色体)

Polytene chromosomes, also referred to as giant chromosomes (巨大染色体), are huge, transversely banded ribbons of DNA. Compared with typical interphase chromosomes, they are longer by one hundred times or more and have diameters thousands of times greater. Only Collembola (トビムシ目) and Diptera (双翅類) among the hexapods (六脚類) have polytene chromosomes, which were first described in larval midges in 1881, and in Collembola in 1961. These giant chromosomes are formed in tissues that grow by cellular enlargement rather than by an increase in cell number. In contrast to a typical mitotic cycle, the homologues of polytene chromosomes typically remain paired and do not participate in the mitotic cycle of coiling and uncoiling. The sister chromatids remain paired at the end of each replication cycle, and the nuclear membrane and nucleoli remain intact throughout replication. 双翅目(ハエ目、Diptera)

唾腺染色体(だせんせんしょくたい, salivary gland chromosome)

唾液腺の細胞にある染色体です(Figure 1)。全ての染色体が動原体(= kinetochore, ≈ セントロメア)のまわりにあるヘテロクロマチンで結合して、染色中心(chromocenter)を成しています。また、相同染色体はそれぞれ、相同な領域同士で対合しています。これらの結果、唾腺染色体は五本の腕(第一染色体, 第二染色体左腕, 第二染色体右腕, 第三染色体左腕, 第三染色体右腕)をもった構造となります(六本目として、小さな第四染色体が見えることもあります)。(ショウジョウバエ入門

唾腺染色体観察の意義、面白さ

  1. 生物学では観察したいことを最も良く観察できる実験材料を選ぶことが研究を成功させる大きな鍵になる。一般的な生命現象を、特殊な実験材料を選ぶことによりその時代の限られた観察技術を用いたとしても容易に観察できるようにするというのは、科学研究を行うときの常套手段。
  2. 唾腺染色体は巨大なので非常に観察しやすい。
  3. 遺伝子の存在(位置)が染色体上のバンドの位置と対応付けらるので、遺伝子の並びを目で見ていることになる!
  4. 転写されている領域がパフとして観察されるので、遺伝子が働いている様子を目で見ていることになる!

Polytene chromosomes from larval salivary glands of D. melanogaster

“Classic” polytene chromosomes from larval salivary glands of D. melanogaster are composed of bundles of one to two thousand tightly synapsed chromosomal strands, which are formed via multiple rounds of endoreplication of just two starting chromatids. As all the homologous chromomeres from all chromatids are aligned to each other, this results in the formation of a thick “cable” with transverse stripes of compacted chromatin (bands) alternating with decompacted interchromomeric regions (interbands). Local differences in size and compaction of banded material form a unique banding pattern that can be used to accurately map any polytene chromosome region. This in turn allows one to link a particular DNA sequence, genes and proteins to the specific chromosomal region, and so to spatially analyze the genetic processes taking place in the interphase nucleus (for review: [1]). 唾腺染色体のバンドの数はいくつあるのか?

The reference system proposed by Bridges divides the limbs of salivary gland chromosomes into 102 sections called “divisions” designated by number from 1 to 102. Each of the five main limbs (X, 2L, 2R, 3L, and 3R) contains 20 divisions; the short chromosome 4 contains only two divisions. The divisions are started with a prominent band and divided further into 6 subdivisions, each designated with capital letters from A to F. Each subdivision starts with a sharp band. Thus each individual band of salivary gland chromosomes can be identified by giving the division number, subdivision, and the number of the band starting from the beginning of the subdivision. Bridges presents the following minimum numbers of bands for the salivary gland chromosomes of Drosophila melanogaster: 537 bands for the X chromosome, 1032 bands for the second chromosome, 1047 bands for the third chromosome, and 34 bands for the fourth chromosome, totalling a minimum of 2650 bands for the whole genome.
